Ciliopathies represent a group of inherited rare diseases due to a defective primary cilium, a ubiquitously expressed organelle of the eukaryotic cells. The primary cilium plays major biological functions in signal capture and transduction. One emblematic ciliopathy, namely the Bardet Biedl syndrome (BBS ) is characterized, among other dysfunctions, by obesity with low diabetic risk . Historically, the obese phenotype in the ciliopathies has been conferred to a hypothalamic central defect in leptin detection causing an induced leptin resistance mediated hyperphagia. Precursors of the adipocytes were shown to be transiently ciliated and that in vitro and in vivo BBS10 or BBS12 inactivation were impacting the adipogenic process favoring increase adipose tissue mass without promoting insulin resistance and diabetes. This post offers a unique opportunity to discover regulatory mechanisms linking ciliary BBS proteins with obesity and insulin resistance through the use of genetically modified mouse models namely Bbs12-/- and Bbs10 floxed mice. In this project we will try to understand: what are the contributions of the hypothalamic ciliary defect versus the impaired adipogenesis process in the development of the obese phenotype in the BBS and how a ciliary defect induced upon BBS inactivation impact insulin sensitivity
To drive this research we are now looking for an enthusiastic and broad thinking individual with a strong background in metabolism, endocrinology or cell biology.
The postdoctoral fellow will be involved in:
1. Working with mice colonies and managing the mice breedings with Cre-expressing mouse lines to ensure specific inactivation of the targeted Bbs allele.
2. Performing the phenotyping of the different mouse lines using a wide range of cellular and molecular assays and analyze the results
Essential Skills
PhD in a relevant subject area, such as metabolism or insulin-sensitive tissues cell biology or endocrinology
Significant skills, knowledge and experience in molecular and stem cell biology
Significant skills, knowledge and experience in genetically modified mouse models and mouse anatomy
A strong publication record in the relevant areas
Highly motivated and independent individual
Ideal Skills
Experience with rodent models of obesity and diabetes
Experience in studying ciliopathies
Postdoctoral Fellows are typically in a period of early career research training (typically their first or second postdoctoral position). This is up to 3 year fixed-term contract with a salary of 2500 euros bruto per month. The position will be based at the Laboratory of Medical Genetics in the Medical school of Strasbourg University, France.
