SECRETARIA REGIONAL DA EDUCAÇÃO E CIÊNCIA
FUNDO REGIONAL DA CIÊNCIA E TECNOLOGIA
ANNOUNCEMENT
SELECTION PROCEDURE M112/FRCT/2007 – SCHOLARSHIP NUMBER 12
EXTENT AND DEADLINE
1 – According with the Portuguese Law nº40/2004, dated August 18th, and with the General Regulation for Scientific Research Scholarships and Management Support of the Science and Technology Regional Fund (Regulamento Geral de Bolsas de Investigação Científica e de Apoio à Gestão do Fundo Regional da Ciência e Tecnologia), available in the Regional Government of the Azores Internet site (http://sctr.azores.gov.pt), we hereby announce that until September 30th, 2007, applications can be submitted for the attribution of a post-doctorate scholarship..
2 – The post-doctorate scholarship derives from the Measure 1.1.2/FRCT – Reinforcement of research teams for accredited R&D Units (Medida 1.1.2/FRCT – Reforço das equipas de investigação das Unidades de I&D acreditadas), funded by the Science and Technology Regional Fund in the frame of the Science and Technology Integrated Plan (Plano Integrado para a Ciência e Tecnologia).
PROJECT
3 – The scholarship aims the development of the project “Aging and susceptibility to infections”.
4 – The main goal of this project is to find an answer to the following question – why oldest organisms are more susceptible to infections than youngest? This is a core question nowadays, when life expectance is increasing continuously world wide and easier communications expose people to new infectious agents. In fact, old people generally are more susceptible to infections, react badly to actual pharmaceuticals, and have a higher rate of adverse reactions to usual therapies than young people. It is clear that the immune system becomes senescent with age however those modifications are not sufficient to explain the highest susceptibility to infections of older persons. To answer this challenge is mandatory to understand basic mechanisms involved in aging process. Many of these studies can not be accomplished with success in humans so animal models must be used.
Several authors studied worms for this purpose. In CIRN laboratories we are studding a worm, which has the specificity of being associated with an entomopathogenic bacteria. We are using the complex nematode-bacteria to control insect pests and to study molecular mechanism of pathogenicity. We perform proteogenomic studies on translational factors has expressed gene analysis using Northernblot, differential dysplay, RT-PCR, cDNA cloning and sequencing, and we are doing siRNA and microinjection. Concerning proteomics we perform 2DE and FPLC enrichment and we have a protocol with ITQB for MALDI-TOF e LC-MS/MS facilities and with CNC for recombinant expression of identified cDNAs. Moreover we perform protein-protein interactions. Bioinformatics is also being used for proteogenomics analysis.
As result of the developed research several tools can be used in this project. From the nematode we have selected strains with high and short longevity, we have genetic high relevant information on this organisms including cDNA libraries of both strains, and from old and aged organisms, protein profiles of these same organisms under different physiological situations including juveniles with higher and lower resistance to bacteria, sequenced mtDNA, etc. We have also a DNA library of the associated bacteria. This complex nematode / bacteria is presently used as a model to study human infections in medicine research laboratories with success.
5 – The work will be developed at the CIRN (http://www.cirn.uac.pt/) , in the University of Azores, based in São Miguel Island, Azores, Portugal under the coordination of Professor Doutor Nelson Simões (simoes@notes.uac.pt).
