Chemical imaging for biomedicine by Coherent Anti-Stokes RamanSpectroscopy (CARS)

Optical microscopy is one of the most widely applied scientific tools,but in biology usually requires a fluorescent agent to give chemicalcontrast. Raman spectroscopy gives chemical information in opticalmicroscopy, as each molecule has a specific vibrational spectrum.However, for molecular species this Raman signal is extremely weak, soimages take many hours to acquire and contrast is poor. CARS is a newvariant of Raman spectroscopy using two lasers to excite molecules, andis 5 orders of magnitude more efficient – so images are acquired inseconds. The system has been installed and has produced CARS images, andis now ready to apply to biomedical imaging.

The successful candidate will optimize the imaging conditions for livecells and tissue, and apply the technique to solve important biologicalproblems – leading to high impact publications. The confocal system alsoincorporates an atomic force microscope, so chemical imaging can beperformed at 10 nm resolution. The microscope also acquiresfluorescence, FLIM, SHG and DIC images.

The candidate should have a track record of advanced optical imaging inbiology, so expertise in SHG, FLIM, multi-photon, Raman or CARS imagingis essential. Experience with atomic force microscopy or pulsed lasersystems, is useful but not essential.

The opportunity to undertake international travel to collaboratinginstitutes and for conference presentations will be available.
