Five year fixed term post on Grade 7: Salary £28,839–£35,469 pa (with discretionary points to £38,757)

Generation of improved transgenic rodent models of Parkinson’s Disease

Applications are invited for a Postdoctoral Career Development Fellow to join the new multidisciplinary initiative, the Oxford Parkinson’s Disease Centre (OPDC) and undertake a highly collaborative and interdisciplinary project to develop improved transgenic rodent models of PD. The Fellow will be based in the Laboratory of Molecular Neurodegeneration in the Department of Physiology, Anatomy and Genetics, and will be supervised by Dr Richard Wade-Martins.

The Fellow will develop improve transgenic rodent (mice and rats) models of PD using bacterial artificial chromosome (BAC)-based transgenesis using wild-type and recombinant variants of genes previously identified within the OPDC as potential candidates underlying susceptibility to PD. BAC transgenics represent the state-of-the-art in transgenic mouse models as the correct temporal and spatial regulation of gene expression is retained in vivo. S/he will use homologous recombination in E. coli to generate mutant BAC constructs expressing PD-related genes. The vectors will be used to generate founder lines by pronuclear injection by Core Transgenic Services. The Fellow will be supported in this work by a full-time Research Assistant responsible for the management of the transgenic colonies. Lines with expression levels closely recapitulating those seen in human brain tissue will be taken forward for phenotypic analysis. We will use comparative gene expression profiling between neuronal subtypes in a healthy or disease state to provide important biological insights into molecular mechanisms of neurological diseases in affected neuronal populations. Finally, there will be opportunity to work closely with other members of the OPDC and have the opportunity to undertake further collaborative studies into the molecular pathology, behavioural phenotype and neuropharmacology of the new models generated. This Fellowship offers the opportunity to train in modern techniques in molecular neuroscience in the highly interdisciplinary research environment of the OPDC.

Applicants must have a PhD or equivalent, post-doctoral research experience in a relevant area of research, a good working knowledge of molecular and cellular biology, experience of working with mice or rats, enthusiasm for scientific research, and an interest in pre-clinical in vitro and in vivo models of neurological disease. A good publication record would be an advantage.
