Description

Regulation of gene transcription in megakaryocytes

Location: University of Cambridge, Department of Haematology, Cambridge, UK

The post is based at the Department of Haematology but will involve close collaboration with the European Bioinformatics Institute and the Wellcome Trust Sanger Institute on the Genome Campus. We seek to appoint an experienced postdoctoral fellow who will work closely with and supervise a number of PhD fellows. The successful applicant will focus on megakaryocyte differentiation with the aim to better define the gene regulatory networks that are operational in this cell over time. This project builds upon the leading role of the Department of Haematology in studies on gene regulation in haematopoietic stem cells. Studies will include over-expression and silencing of target genes by lentiviral-mediated modification of haematopoietic stem cells and subsequent differentiation into megakaryocytes. Subsequent effects on gene expression will be determined using whole genome expression arrays. ChIP-Seq experiments on key DNA binding proteins will allow the development of Bayesian transcription factor networks of regulation of gene transcription. These networks will be informed by transcription factor pull-down from megakaryocytes that will reveal relevant protein-protein interactions (collaboration with University College Dublin).

The ideal candidate will have a PhD in molecular and cell biology, ideally with experience in the isolation, culture and differentiation of CD34+ haematopoietic stem cells. Previous experience of transcription factor networks is desirable.
